Tetramethyl orthosilicate (also named as Tetramethoxysilane,TMOS) is popularly used for the synthesis of quartz, high-purity silica and sol-gel. It is also used as ceramics binder. Most recently, TMOS has been explored as an novel reagent for the direct amidation of aliphatic and aromatic carboxylic acids with amines and anilines. A variety of amide products can be prepared by combining a carboxylic acid react with an amine in refluxing toluene in the presence of TMOS as a dehydration-condensation agent to provide the corresponding amides in good yields. This reaction can proceed amino acids as well as general carboxylic acids and amines. Furthermore, this method is expected to be used in the scale-up synthesis of organic materials and pharmaceuticals due to the direct method and excellent atom economy.
